Last years clinical and experimental research has shown that the serotoninergic system is involved in the pathogenesis of obsessive-compulsive disorder. Clomipramine and SSRI have already shown their efficacy in the treatment of OCD, while more recent studies show the potential efficacy of venlafaxine. Our study is aimed at evaluating the efficacy of venlafaxine in patients affected by OCD. Method: 19 patients who satisfy DSM-IV-TR criteria for OCD were enrolled. 11 of these completed the 3 months follow up period. They were treated with venlafaxine (mean dose: 167 mgs per day; range 37,5-300 mgs per day). We used the Yale-Brown Obsessive-Compulsive Scale (Y-BOCS) and the Hamilton Rating Scale for Depression (HAM-D) to assess their symptoms. These scales were used to assess the patients before starting the treatment (T0), and after the first (T1) and the third (T2) month of treatment. Results: 55% of the patients responded to the treatment with venlafaxine. Response is defined as a 35% reduction of the initial score of the Yale-Brown Obsessive Compulsive Scale (Y-BOCS). The mean reduction of the score obtained in both scales (Y-BOCS and HAM-D) has been statistically significant (respectively p < 0.0028 and p < 0.01). Significant predicting factors of clinical efficacy have not been registered. The treatment was efficacious independent of anagraphical (gender, age) characteristics, the dose, the presence of comorbidity with other disorders or the unresponsiveness to previous treatments with SSRI. Conclusions: venlafaxine seems to be efficacious both on obsessive-compulsive and depressive symptoms associated to OCD.[:]
